A user control is registered on the top of the Default.aspx page and it is used .

are you sure its because of the user control,did you try removing that user control?

If you include the code snippet, in which the core content is written, chances of getting replies will be more.

Process cannot access the file, because it is being used by another process could be due to several reasons.
1) Check whether the file is exclusively opened by you or any other user.
2) Check whether the file is not closed using your code.
