Running one application in multiple systems

generally ASP.NET, IIS and SQL Server handle all such issues internally i.e. simulatenous updating of database. Unless, a really fine grained control over the timing and sequencing of operations is needed in which case you would have to lock the resource being updated so that no one else can change it during that time. Unfortunately, thats all I can say from the limited information present in your question.
