If I have understood you correctly, you just have to skip first two strings in concatenation ('/' + 'new value') which will leave you this:
SUBSTRING(ReportLocation, CHARINDEX(ReportLocation, '/', 2), LEN(ReportLocation)).

What have you tried so far to detect problem source?
I suggest you to invest some time to learn and implement some logging mechanism, without it you are virtually blind.
Check EventViewer and IIS logs, that's all I can do for you unless you provide more information.

Lately I only use custom methods and Entity framework, but I'll never say they're best choice.
I have no professional experience in LINQ to SQL, but I doubt I'll ever try to use it. It's OK technology, but it's not evolving.
My choice of technology depends on data source, client's demands and how soon it must be done.

It rarely returns more than 35k rows. Schema is datetime2, int, int, int

DataGridView with its dataSource, dataSource can automatically add, remove and update data; I change data with Command buttons on grid.

Updating data works well, deleting also, when I navigate off and on the page data shown in grid is up-to-date, my problem only occurs when I am trying to refresh a page and I wonder why it happens?

It never happened with SQLDataSource or ObjectDataSource.

1. There is no mention of your ddl in code behind you submitted here.
Change that and perhaps someone will be able to help.
2. Ddl from your markup is called ddl_formstatus and ddl from your error message is dropdownlist1, therefore I think it is possible that code you have submitted here isn't source of your error.
